    Human mesenchymal stromal cells from different sources diverge in their expression of cell surface proteins and display distinct differentiation patterns

    When germ-free cell cultures became a laboratory routine, hopes were high for using this novel technology for treatment of diseases or replacement of cells in patients suffering from injury, inflammation, or cancer or even refreshing cells in the elderly. Today, more than 50 years after the first successful bone marrow transplantation, clinical application of hematopoietic stem cells is a routine procedure, saving the lives of many every day. However, transplanting other than hematopoietic stem and progenitor cells is still limited to a few applications, and it mainly applies to mesenchymal stromal cells (MSCs) isolated from bone marrow. But research progressed and different trials explore the clinical potential of human MSCs isolated from bone marrow but also from other tissues including adipose tissue. Recently, MSCs isolated from bone marrow (bmMSCs) were shown to be a blend of distinct cells and MSCs isolated from different tissues show besides some common features also some significant differences. This includes the expression of distinct antigens on subsets of MSCs, which was utilized recently to define and separate functionally different subsets from bulk MSCs. We therefore briefly discuss differences found in subsets of human bmMSCs and in MSCs isolated from some other sources and touch upon how this could be utilized for cell-based therapies

    Buckling of elastically restrained nonlocal carbon nanotubes under concentrated and uniformly distributed axial loads

    Buckling of elastically restrained carbon nanotubes is studied subject to a combination of uniformly distributed and concentrated compressive loads. Governing equations are based on the nonlocal model of carbon nanotubes. Weak formulation of the problem is formulated and the Rayleigh quotients are obtained for distributed and concentrated axial loads. Numerical solutions are obtained by Rayleigh–Ritz method using orthogonal Chebyshev polynomials. The method of solution is verified by checking against results available in the literature. The effect of the elastic restraints on the buckling load is studied by counter plots in term of small-scale parameter and the spring constants.</p

    Variational Principles for Multiwalled Carbon Nanotubes Undergoing Vibrations Based on Nonlocal Timoshenko Beam Theory

    Variational principles are derived for multiwalled carbon nanotubes undergoing linear vibrations using the semi-inverse method with the governing equations based on nonlocal Timoshenko beam theory which takes small scale effects and shear deformation into account. Physical models based on the nonlocal theory approximate the nanoscale phenomenon more accurately than the local theories by taking small scale phenomenon into account. Variational formulation is used to derive the natural and geometric boundary conditions which give a set of coupled boundary conditions in the case of free boundaries which become uncoupled in the case of the local theory. Hamilton's principle applicable to this case is also given

    Detecting user types in object ranking decisions

    With the emergence of Web 2.0 applications, where information is not only shared across the internet, but also syndicated, evaluated, selected, recombined, edited, etc., quality emergence by collaborative effort from many users becomes crucial. However, users may have low expertise, subjective views, or competitive goals. Therefore, we need to identify cooperative users with strong expertise and high objectivity. As a first step towards this aim, we propose criteria for user type classification based on prior work in psychology and derived from observations in Web 2.0. We devise a statistical model for many different user types, and detection methods for those user types. Finally, we evaluate and demonstrate both model and detection methods by means of an experimental setup. Copyright 2009 ACM

    The advanced video information system: Data structures and query processing

    We describe how video data can be organized and structured so as to facilitate efficient querying. We develop a formal model for video data and show how spatial data structures, suitably modified, provide an elegant way of storing such data. We develop algorithms to process various kinds of video queries and show that, in most cases, the complexity of these algorithms is linear. A prototype system, called the Advanced Video Information System (AVIS), based on these concepts, has been designed at the University of Maryland

    Optimization and analysis of frequencies of multi-scale graphene/fibre reinforced nanocomposite laminates with non-uniform distributions of reinforcements

    Optimal design and analysis of three-phase graphene/fibre reinforced laminated nanocomposite plates with respect to maximizing the fundamental frequency is the subject of the present study. Optimal design solutions are given for four different sets of design parameters. First design problem determines the optimal graphene contents of individual layers, the second one both graphene and fibre contents, the third optimizes the graphene and fibre contents as well as the layer thicknesses of individual layers, and the fourth problem optimizes the graphene and fibre contents, layer thicknesses and fibre orientations. Purpose of this approach is to assess and compare different levels of optimization by means of a design efficiency index and as such to determine the effectiveness of different design parameters in maximizing the fundamental frequency. Optimization is implemented using a Sequential Quadratic Programming algorithm and the mechanical properties of graphene/fibre nanocomposite are determined via micromechanical relations. Vibration analysis is conducted by the finite element method using four-noded Mindlin plate elements. Results are obtained for simply supported (SSSS), clamped (CCCC) and simply supported-clamped boundary conditions for opposite edges (SCSC). It is observed that non-uniform distributions of graphene and fibre as well as fibre orientations are quite effective in improving the design efficiency

    Epilobium hirsutum alters xenobiotic metabolizing CYP1A1, CYP2E1, NQO1 and GPx activities, mRNA and protein levels in rats

    Context: Natural products have attracted increasing interests due to their use in flavoring, nutrition, cosmetics, pharmacy and medicine. Epilobium hirsutum L. (Onagraceae) is known for its analgesic, antimicrobial, and antiproliferative activity. CYP1A1 and CYP2E1, xenobiotic metabolizing enzymes, serve as a metabolic activation route yielding reactive metabolites that are eliminated by the action of NQO1 and glutathione peroxidase (GPx) enzymes. Objective: This study investigated in vivo effects of Epilobium hirsutum (EH) on CYP2E1, CYP1A1, NQO1 and GPx activities, protein and mRNA expressions in liver. Materials and methods: Male Wistar Albino rats were injected with EH at a dose of 37.5mg/kg i.p. daily for 9d. CYP2E1, CYP1A1, NQO1 and GPx activities, protein and mRNA levels were determined by enzyme assays, Western blotting and qPCR, respectively. Results: CYP1A1 associated ethoxyresorufin-O-deethylase activity of control and EH-treated animals were found as 6.54±1.21 and 4.48±1.67nmol/min/mg, respectively. CYP2E1 associated aniline 4-hydroxylase of control and EH group were 0.537±0.011 and 0.109±0.01nmol/min/mg, respectively. However, EH treatment increased the GPx and NQO1 activities from 0.069±0.015 to 0.107±0.026nmol/min/mg and from 163.34±92 to 588.3±14nmol/min/mg, respectively. Furthermore, protein and mRNA expression analysis revealed that CYP1A1 and CYP2E1 levels were decreased while those of NQO1 and GPx increased after EH treatment. Discussion and conclusion: Our current data suggest that the metabolism of xenobiotics, including drugs, may be altered due to changes in the expression and activity of these proteins by EH. © 2013 Informa Healthcare USA, Inc

    An MPEG-7 scheme for semantic content modelling and filtering of digital video

    Abstract Part 5 of the MPEG-7 standard specifies Multimedia Description Schemes (MDS); that is, the format multimedia content models should conform to in order to ensure interoperability across multiple platforms and applications. However, the standard does not specify how the content or the associated model may be filtered. This paper proposes an MPEG-7 scheme which can be deployed for digital video content modelling and filtering. The proposed scheme, COSMOS-7, produces rich and multi-faceted semantic content models and supports a content-based filtering approach that only analyses content relating directly to the preferred content requirements of the user.
